Thymic T-Cell Education: Positive & Negative Selection

ثبت نشده
چکیده

TOLERANCE (which is antigen-specific) must be distinguished from immune deficiency (non-specific). Its most important manifestation, the maintenance of SELFTOLERANCE, was originally explained by the Clonal Selection theory as the result of CLONAL ABORTION of self-reactive clones. However, potentially self-reactive Tand B-cells do exist in normal individuals, leading to the recognition of the importance of active processes in the maintenance of tolerance, such as SUPPRESSION. The dynamic and competing balance between immunity and tolerance is discussed, together with three useful model systems, INDUCED CLONAL ABORTION, RECEPTOR BLOCKADE, and ANTIGEN SEQUESTRATION. Two general categories of tolerance are recognized, CENTRAL TOLERANCE which is generated during development of T-cells within the thymus, and PERIPHERAL TOLERANCE generated by a variety of mechanisms outside this central lymphoid organ.

The thymic repertoire of neuroendocrine-related self antigens: biological role in T-cell selection and pharmacological implications.

Thymic epithelium, including nurse cells (TEC/TNC), as well as other thymic stromal cells (macrophages and dentritic cells), express a repertoire of polypeptide belonging to various neuroendocrine protein families (such as the neurophypophysial, tachykinin, neurotensin and insulin families). A hierarchy of dominance exists in the organization of the thymic repertoire of neuroendocrine precursor...

Bim-mediated apoptosis is not necessary for thymic negative selection to ubiquitous self-antigens.

T cell education in the thymus is critical for establishing a functional, yet self-tolerant, T cell repertoire. Negative selection is a key process in enforcing self-tolerance. There are many questions that surround the mechanism of negative selection, but it is currently held that apoptosis initiated by Bim and/or Nur77 is critical for negative selection. Recent studies, however, have question...

Clonal deletion induced by either radioresistant thymic host cells or lymphohemopoietic donor cells at different stages of class I-restricted T cell ontogeny

Major histocompatibility complex (MHC) products and self-antigens expressed in the thymus determine the repertoire of mature alpha/beta T cells. While positive selection of self-MHC-restricted T cells is directed by MHC molecules expressed by thymic epithelial cells, negative selection depends to a large extent on self-antigens presented by lymphohemopoietic cells. However, radioresistant compo...

TGF-beta signalling contributes to thymic epithelial cell damage and regeneration following myeloablative conditioning and stem-cell transplantation

Hematopoietic stem cell transplantation (HSCT) is considered a potentially curative therapy for a number of severe autoimmune diseases. The rationale consists of deleting auto-reactive T and/or B cell clones and re-constructing a functional immune system tolerant to autoantigens. The re-emergence of T cell immunity following myeloablation largely depends on thymic de novo production of naïve T ...

Dissociation of thymic positive and negative selection in transgenic mice expressing major histocompatibility complex class I molecules exclusively on thymic cortical epithelial cells.

Thymic positive and negative selection of developing T lymphocytes confronts us with a paradox: How can a T-cell antigen receptor (TCR)-major histocompatibility complex (MHC)/peptide interaction in the former process lead to transduction of signals allowing for cell survival and in the latter induce programmed cell death or a hyporesponsive state known as anergy? One of the hypotheses put forwa...

Mutation of the phospholipase C-γ1–binding site of LAT affects both positive and negative thymocyte selection

Linker for activation of T cells (LAT) is a scaffolding adaptor protein that is critical for T cell development and function. A mutation of LAT (Y136F) that disrupts phospholipase C-gamma1 activation and subsequent calcium influx causes a partial block in T cell development and leads to a severe lymphoproliferative disease in homozygous knock-in mice. One possible contribution to the fatal dise...
